چکیده

The core histones have long been known to have an important role in the packaging of DNA into nucleosomes. Today, nucleosome assembly is recognized as an integral component of both transcriptional activation and repression [1]. Transcriptional control is mediated through the assembly of extensive nucleoprotein structures containing both conventional transcription factors and architectural factors [2]. The latter include high mobility group proteins (HMGs), histones and proteins of similar structure that direct the folding of DNA necessary for the appropriate assembly of regulatory complexes. Recent progress has determined that the inclusion of histones into regulatory complexes depends upon specific protein–protein interactions with trans-acting regulatory factors [3,4]. These observations indicate that an intimate relationship exists between regulatory proteins and the histones, both within and outside the nucleosome.

STU DIES ON THE BINDING OF THE ALKYLATING AGENT SULFUR MUSTARD TO CALF THYMUS CHROMATIN

In this study the effect of the alkylating agent, sulfur mustard, on calf thymus chromatin was investigated using UV/Vis spectroscopy, gel electrophoresis and thermal denaturation techniques. The results show that treatment of isolated chromatin with sulfur mustard releases histones from the core particles but does not affect histone H I and nonhistone chromosomal proteins. The content of ...

DNA methylation of tumor suppressor genes in hepatocellular carcinoma

The basic unit of chromatin is a nucleosome included an octamer of the four core histones and 147 base pairs of DNA. Posttranslational histones modifications affect chromatin structure resulting in gene expression changes. CpG islands hypermethylation within the gene promoter regions and the deacetylation of histone proteins are the most common epigenetic modifications. The aberrant patterns of...

Post-translational changes of histones, methylation level, and ERβ protein level in the cumulus cell genome of infertile women with endometriosis

Background: Endometriosis (which affects up to 50% of infertile women) is one of the major causes impacting female infertility. Endometriosis, defined as the presence of endometrial glands and stroma outside the uterine tissue, causes a wide range of functional disorders in the process of follicular development and changes in the follicular milieu, resulting in the formation of an incompetent o...

I-40: Male Genome Programming, Infertility and Cancer

Background: During male germ cells differentiation, genomewide re-organizations and highly specific programming of the male genome occur. These changes not only include the large-scale meiotic shuffling of genes, taking place in spermatocytes, but also a complete “re-packaging” of the male genome in post meiotic cells, leading to a highly compacted nucleo-protamine structure in the mature sperm...

Fractionation of liver chromatin.

Rat liver chromatin has been fractionated into two fractions on the basis of precipitability in standard saline after mild treatment with DNase II. The major portion of liver chromatin contains little nonhistone protein and is enriched in histones, while a minor portion of such chromatin, with which RNA polymerase is associated, is highly enriched in proteins other than histone and impoverished...
